A phenotypic HTS for the indication osteoarthritis testing more than 500 000 compounds for differentiation and proliferation of primary pig chondrocytes will be described. Details will be given on cell selection and cell logistics for large scale screening. Assay technology, readout, statistics and results from the HTS screen will be presented as well.
In a second part, the assay cascade for validation of the primary hits will be discussed, concentrating on the applicability of hanging drop 3Dcultures.
