This talk will summarise the electrophysiological properties of iPSC-derived cardiomyocytes as assessed using voltage sensitive dyes on the CellOPTIQ platform (Clyde Biosciences). This system provides a ratiometric assessment of membrane potential from the fluorescence of Di-4-ANEPPS in iPSC-CMs seeded on a 96 well plate format. Commercial sources of iPSC-CMs have different electrophysiological baseline characteristics and appear to respond differently to rapid stimulation. But restitution analysis indicates a similar relationship between diastolic interval and action potential duration. Cells from different sources respond differently to drugs targeted to different ion channels, but demonstrate effects consistent with their known targets with a sensitivity similar to human adult myocardium.
