We will compare HTS methodologies used to screen membrane bound PPI targets, including techniques currently employed by UCB in early drug discovery programs.

Tool Peptides will be used to facilitate direct comparisons between the sensitivity of different Biochemical and Cellular competition assays, including FRET, HTF and FMAT. Data from assay validation experiments will be used to compare precision, the potential incidences of false positives and the subsequent thresholds for Hit ID.
In addition, we will consider the ability of methods to identify chemical inhibitors with allosteric mechanisms of action.
