Abstract

Multiplexed assays allow harvesting more information from each sample and can accelerate the pace of drug discovery. FirePlex® multiplexed assay technology is based on fluorescent dye encoded micro-particles and an intuitive decoding workbench software. FirePlex® allows measuring up to 65 miRNAs or 70 proteins in a single well of a 96-well plate. The fluorescence signal is acquired using a flow-cytometer. For higher throughput, FirePlex® has been adapted to interface with laboratory automation using a 384-well plate format and a no-wash assay workflow. Ten protein analytes can be simultaneously quantified in the miniaturized assay format. The signal in this case is acquired using a high-content imager. The content of the immunoassays can be easily customized by mixing and matching particles and assay reagents for various targets and thus allow focusing on distinct research areas. This versatile multiplexed assay technology is ideal for cell-line characterization and can help uncover new cellular phenotypes and speed-up the development of new in-vitro model systems. Specific examples will be provided.
